How does defensive fire work in the game? Which units can do this skill? And if a unit has ''cover/supressing fire'' skill how does that effect it.

I'm a little unsure how to answer this. If you've read the rules, how can I be more clear than that? And if you haven't, why not?

Rulebook, p. 21: Provoking Defensive Fire
A unit provokes defensive fire when it moves from one hex adjacent to a unit into another hex adjacent to that same enemy unit. The hex a unit is in counts as adjacent to that unit.

What's unclear about that? All units can make one defensive fire per phase, unless special circumstances or abilities restrict it. But there are at least a couple of special situations. One of them is the Suppressive Fire special ability, which allows a unit with this ability to make an unlimited number of defensive fires (one per triggering event).

Another is in the rulebook just after the section I quoted above, under the heading Soldiers and Vehicles: Soldiers don't provoke defensive fire from Vehicles.

Yes. The only exception in the RAW for defensive fire is tanks against soldiers. IIRC, the Overlapping Fields of Fire SA grants a tank with that ability defensive fire against soldiers; otherwise, no.
