For example if you have 6 infantry that roll 2 dice each in close combat against a tank (rear attack 3). You roll 12 dice or 2 dice 6 times?

If the answer is 2 dice 6 times this means that the six infantry can never destroy that tank (armor 3 and the maximum attack of those infantry is 2). Is that correct?

No attacks stack with any other attacks. Every fistfull of dice is examined separately to see how many successes and hits it achieves.

Your example is correct if the infantry are at 1-hex range from the tank. With only two dice each, they cannot hurt a vehicle with defense 3 or higher, no matter how many of them attack. If they move into the tank's hex, however, their attacks become much more effective because they can use their Close Assault values instead of their normal anti-tank values.
