If a unit with the flamethrower capability fires during Defensive Fire and rolls 3 Sixes on the dice, does the unit being fired at get destroyed?
In the rules it states that the most that can happen from Defensive fire is a disruption counter regardless of the amount of successes. Then the card says something different.

In our store we have played the destroyed result.

Special abilities take precedence over the standard rules. In effect, they allow a unit to 'break the rules'. So yes, a flamethrower using defensive fire destroys the target immediately on three 6s.
